Abstract

¿¬±¸ ¸ñÀû: º» ½ÇÇèÀº MS275¿Í PLGA (poly lactic-co-glycolic acid) ÀÇ º¹ÇÕü¸¦ submicron Å©±â·Î ƼŸ´½ µð½ºÅ© Ç¥¸é¿¡ ÄÚÆÃÇÏ¿©, Ç¥¸éÀÇ º¯È­¸¦ ¾Ë¾Æº¸°í »ý¹°ÇÐÀûÀ¸·Î °£¿± Áٱ⼼Æ÷ È°¼º¿¡ ¾î¶°ÇÑ ¿µÇâÀ» ¹ÌÄ¡´ÂÁö Á¶»çÇϱâ À§ÇØ ½ÃÇàµÇ¾ú´Ù.

°á°ú: AFM (atomic force microscope) °á°ú ´ëÁ¶±º°ú ½ÇÇ豺¿¡¼­ °ÅÄ¥±âÀÇ À¯ÀÇÇÒ¸¸ÇÑ Â÷ÀÌ°¡ ¾ø¾ú´Ù(P>.05). MTT °á°ú 1, 4, 7ÀÏ ½Ã°£ÀÌ Áö³²¿¡ µû¶ó ¼¼Æ÷ Áõ½ÄÀÌ È°¹ßÇØÁ³À¸¸ç ¼¼Æ÷¹è¾ç 7ÀÏ¿¡ MS275 ³óµµ ¾È¿¡¼­, MS275ÀÇ ³óµµ°¡ Ä¿Áü¿¡ µû¶ó ¼¼Æ÷ÀÇ È°¼ºµµ°¡ ³ô¾ÆÁüÀÌ À¯ÀÇÇÒ ¼öÁØÀ¸·Î È®ÀεǾú´Ù(P<.05). ¼¼Æ÷ ºÎÂøÀ» SEMÀ¸·Î È®ÀÎÇÑ °á°ú, ¼¼Æ÷ÀÇ ºÎÂø ¼ö´Â ½Ã°£ÀÌ °¥¼ö·Ï Áõ°¡ÇÏ°í ºÎÂø ÇüÅ ¿ª½Ã µ¹±â°¡ Å©°í ³Ð¾îÁö¸ç, Ç¥¸é°ú ±ä¹ÐÇÔ Á¢ÃËÀÌ Áõ°¡ÇÏ¿´´Ù.

°á·Ð: FE-SEM°ú MTT °á°ú MS275/PLGA º¹ÇÕü·Î Ç¥¸é ó¸®µÈ ŸÀÌŸ´½ Ç¥¸é¿¡¼­ ¼¼Æ÷°¡ Ãʱ⿡ (7Àϳ») ºü¸£°Ô Áõ½ÄÇÏ¿´´Ù. ¶ÇÇÑ º¹ÇÕü 󸮱ºÀÇ ³óµµ°¡ Áõ°¡ÇÒ¼ö·Ï ³ôÀº ¼¼Æ÷ ¼ºÀå ¼öÄ¡¸¦ º¸¿´´Ù.

Purpose: This study was conducted to identify the surface characteristics of titanium discs coated with MS275/PLGA by electrospray and which is effective to mesenchymal stem cell proliferation.

Materials and Methods: We used anodized surface coated with PLGA as a control group and anodized surface coated with MS275 , , as test groups. To examine that the coating particles are nanometer sized, FE-SEM was used and AFM was utilized to determine the difference of coating surface roughness. We checked the mesenchymal stem cell proliferation by using MTT assay on , , days.

Results: There was no significant difference between control groups and test groups in AFM results (P>.05). In MTT assay results, mesenchymal stem cell proliferation was increased with time, at day, cell viability on discs coated with MS275 was significantly higher than control group (P<.05). As SEM showed, the number of cells on all discs was increased and the morphology of cell attachment was also wider and closer with time.

Conclusion: Titanium surface coated with MS275/PLGA showed significantly higher cell proliferation and the more density of MS275 was dispersed on titanium discs, the faster cells grew.
